Localizing glucose transport proteins: active investigation of passive carriers.

چکیده
glucose transporter: a basolateral protein in hepatocytes and intestine and kidney cells. ABSTRACT The " liver " isoform of the facilitated diffusion glucose transporter is expressed predominantly in liver, intestine, kidney, and pancreatic islet p-cells. The apparent molecular mass of the transporter in liver, kidney, and intestine is different, as detected by Western blot analysis of membrane proteins using antipeptide antibodies. However, as assessed by Northern blot analysis and molecular cloning, the same mRNA is expressed in these tissues, indicating that there are tissue-specific posttranslational modifications of the same transporter polypeptide. As determined by immunofluorescence analysis on frozen tissue sections, the liver glucose transporter is present on the sinusoidal membrane of hepatocytes, on the basolateral membrane of fully differentiated ab-sorptive intestine epithelial cells, and on the baso-lateral membrane of proximal tubule cells of the kidney nephron. This localization is consistent with the involvement of the liver glucose transporter in several key steps of glucose metabolism: glucose uptake and release by the liver and absorption or reabsorption by epithelial cells of the intestine and kidney, respectively. Glucose transporters in cattle - a review*
Glucose is the major energy source for the animal cells. It is important substrate for protein and lipid synthesis. This sugar is absorbed into the cells from body fluids via glucose transporters – structurally related trans-membrane proteins. There are two types of glucose transport passive and active. The facilitative-glucose-transporter family of proteins (solute co-transporters GLUT, encode...
متن کاملStimulation of active and passive sodium absorption by sugars in the human jejunum.
The effects of glucose and fructose on water and sodium absorption in the human jejunum were compared to assess the relative contribution of active and passive sugar stimulation of sodium transport. The effect of fructose is assumed to be entirely passive, and the difference between the effects of fructose and glucose is assumed to be a measure of sugar-stimulated, active sodium absorption. Wat...
متن کاملEffect of vasoactive intestinal polypeptide on active and passive transport in the human jejunum.
The effect of intravenous vasoactive intestinal polypeptide (VIP) on normal transport mechanisms in the human jejunum in vivo was examined with the triple-lumen, steady-state perfusion technique. By using special test solutions that revealed different aspects of jejunal transport, we were able to evaluate the effect of VIP on specific transport processes, such as active bicarbonate absorption, ...
متن کاملHuman adipocyte glucose transport system. Biochemical and functional heterogeneity of hexose carriers.
We have investigated glucose transport proteins in isolated human adipocytes. Using the cytochalasin B binding assay to measure glucose transporters in subcellular membrane subfractions, we found that insulin induced translocation of intracellular glucose transporters to the cell surface. Isoelectric focusing of glucose transporters photolabeled with [3H]cytochalasin B revealed two distinct glu...
متن کاملThe effect of surgical bowel manipulation and anesthesia on intestinal glucose absorption in rats.
The effects of surgical bowel manipulation and anesthesia on intestinal glucose absorption were determined in chronically catheterized rats. Total and passive rates of glucose absorption were measured using 3-O-methyl-glucose (3OMG) and L-glucose, metabolically inert analogues of D-glucose. The rates of 3OMG absorption immediately postoperative and 4 h later were 86 and 62% less than the absorp...
